چکیده

An appearance suggesting extreme fragmentation of chromosomes has been referred to as "pulverization" by Nichols et al. (1). A different appearance, that of accentuated secondary constrictions (or chromomeres), was described by Stubblefield in a Chinese hamster cell line, Don (2). In 1967 zur Hausen (3) described an appearance more often occurring in chromosome No. 1 and referred to as "incomplete condensation." The incompletely condensed segments were shown to synthesize DNA after the normal segments had ceased replication. Subsequently, both Nichols et al. (4) and Kato and Sandberg (5) have reported that pulverized chromosomes exhibit a similar asynchrony. The present report will provide evidence that pulverization, incomplete condensation, as well as some varieties of enhanced secondary constrictions form a spectrum of changes characterized by their spontaneous occurrence in permanent cell lines of several species and by DNA synthesis in G2 .

Chromosome patterns of human leukocyte established cell lines.

Of 14 lines of human leukocytes, designated LK1D, LK57, LK60, LK63, LK87A, LK91, SK-L1, SK-L3, SK-L4, SK-L5, SK-L6, SK-L7, RPMI 7216, and RPMI 7466, all were classi cally diploid or very near-diploid when first examined. All but one showed occasional No. 10 chromosomes with the near terminal secondary constriction seen in Burkitt lymphoma cells in vitro. Nine lines showed accentuation of second...

Locations of 18S and 28S ribosomal genes on the chromosomes of the Indian muntjac

The locations of genes coding for 18S and 28S ribosomal RNA have been mapped on metaphase chromosomes of the Indian muntjac M. muntjak by in situ hybridization with (3H)rRNA from the toad X. laevis. The results show that, in the muntjac, rDNA clusters are associated with the prominent secondary constrictions on the X and the Y1 chromos. In addition a cluster of rDNA is found near the tip of one...

Sequential changes in spleen cell chromosomes during Friend virus leukemia.

Infection of BALB/c mice with varying doses of Friend virus resulted in the appearance of abnormal chromosome numbers in spleen cells. Polyploid cells were found following infection with all doses of virus used. Statistically, however, polyploidy showed no correlation with virus dose, time after infection, or their combined interaction. A total of 0.9% of all infected metaphase figures examined...

Chromosome Pulverization in Human Binucleate Cells following Colcemid Treatment

Under the influence of Colcemid, a substantial number of binucleate human cells from a line infected with herpes-like virus was found to possess pulverized chromosomes. Although this abnormality was also detected in untreated binucleate cells, the increase in the number of pulverized cells after the addition of Colcemid was too striking to be explained by accumulation of spontaneously occurring...

Initiation of DNA synthesis in the prematurely condensed chromosomes of G1 cells

The objective of this study was to investigate whether G1 cells could enter S phase after premature chromosome condensation resulting from fusion with mitotic cells. HeLa cell synchronized in early G1, mid-G1, late G1, and G2 and human diploid fibroblasts synchronized in G0 and G1 phases were separately fused by use of UV-inactivated Sendai virus with mitotic HeLa cells. After cell fusion and p...
